Biomaterial-based cancer vaccine combines chemo and immunotherapy to treat triple-negative breast cancer - Medical Xpress

Image
Dendritic cells, like the one seen here in yellow, pick up antigens from tumor cells and carry them to the lymph nodes and spleen, where they are presented to T cells that mount an immune attack against the tumor. Credit: Wyss Institute at Harvard University Patients with cancer have multiple treatment options available to them today, but each has its drawbacks. Chemotherapy kills rapidly dividing cancer cells, but it also damages healthy cells in the body and often does not effectively prevent tumor metastasis or disease recurrence. Immunotherapies avoid those problems by acting on a patient's immune system to generate a sustained anti-cancer response, but frequently have trouble accessing tumors due to the immunosuppressive local environment that tumors create. Now, a new, best-of-both-worlds approach packages the cancer-killing power of chemotherapy and the long-term efficacy of immunotherapy into a biomaterial-based cancer vaccine that can be injected adjacent to a tumo...

Intravenous SNAP Cancer Vaccine Generates Stem-Like (TCF1+) antigen-Specific CD8+ T cells Resulting in Improved Efficacy Tweet this "Recent discoveries in cancer immunology have demonstrated the importance of TCF1 + CD8 T cells, which have stem-like properties and show enhanced proliferation in response to checkpoint inhibitors," said Dr. Andrew Ishizuka , Avidea's Chief Scientific Officer. "Our findings reported in Nature Immunology show how the design and route of administration of the SNAP Cancer Vaccine allows for control over the proportion of TCF1 + CD8 T cells generated in vivo ." "Many vaccines can only be given by the intramuscular or subcutaneous route. By design, SNAP CV can be administered by any route, including IV, which provides maximal flexibility to control the differentiation of anti-tumor T cells as we pursue SNAP CV monotherapy and combination immunotherapies to combat cancer," said Dr. Geoffrey Lynn , Avidea's Chief Execut...

Cancer Vaccine Plus Keytruda Shows Promise in Advanced Melanoma Trial - Immuno-Oncology News

Image
Adding UV1, Ultimovacs’ experimental cancer vaccine, to Keytruda (pembrolizumab) is safe and shows initial signs of effectiveness in adults with previously untreated metastatic melanoma, according to top-line data from a Phase 1 clinical trial. “The safety profile observed to date in this first cohort [group of patients] of the [UV1-Keytruda] combination study is consistent with the promising safety profiles seen in our earlier Phase 1 trials,” Jens Bjørheim, Ultimovacs’ chief medical officer, said in a press release. “We are encouraged by the results and look forward to additional data readouts over the next years that will demonstrate if the initial signs of efficacy in this cohort persist and mature into long-term added clinical benefit for the patients,” he added. The UV1 vaccine works by promoting immune responses against the human telomerase reverse transcriptase (hTERT), an enzyme normally inactive in healthy cells and active in 85%–90% of all cancer cell typ...

NEW YORK – Results from a Phase Ib clinical trial published Thursday in the journal Cell demonstrated the safety and preliminary efficacy of combined treatment with the personalized, neoantigen-based vaccine NEO-PV-01 and the anti-PD-1 checkpoint inhibitor nivolumab (Bristol Myers Squibb's Opdivo) for patients with advanced melanoma, non-small cell lung cancer, or bladder cancer. BioNTech subsidiary Neon Therapeutics designed NEO-PV-01 to target neoepitopes encoded by somatic mutations in patients' tumors. Each personalized iteration of the vaccine evaluated in this trial consists of up to 20 unique peptides selected using bioinformatics algorithms following whole-exome and RNA sequencing of patients' tumor samples. In the Phase Ib study, patients received nivolumab for 12 weeks while their unique vaccine was manufactured. Patients did not have to meet a PD-L1 expression threshold in order to receive the immunotherapy. Ultimately, 60 patients received NEO-PV-01, includi...

A research team at Oslo University Hospital in Norway has opened a mesothelioma clinical trial using the novel UV1 cancer vaccine alongside a promising immunotherapy combination. UV1 is a peptide-based vaccine designed to induce a specific T-cell response and increase the effectiveness of the immunotherapy drugs. This is the first time UV1 will be studied with mesothelioma cancer, but it already has shown safety and signs of efficacy when used on malignant melanoma, prostate cancer and lung cancer in studies worldwide. It will be used with the immunotherapy combination of nivolumab and ipilimumab, also known by brand names Opdivo and Yervoy, respectively. UV1 is a product of Ultimovacs, a pharmaceutical company based in Oslo, Norway. The study will soon move into six other national hospitals, covering Sweden, Spain, Denmark and Australia. “Even if we can’t promise efficacy at this point, we believe mesothelioma is a relevant cancer in which to test this vaccine,” Dr. Jens Bjorhei...
